CMV IgM Elisa test is intended for the qualitative detection of IgM antibody to CMV in human serum for the presumptive diagnosis of acute, recent, or reactive CMV infection.

Product Description:

CMV IgM refers to the detection of IgM antibodies to cytomegalovirus in a person's blood. IgM antibodies are a specific type of immunoglobulin that is usually produced during a primary infection or when an infection is exacerbated. Therefore, a positive CMV IgM result may indicate a current or recent infection with cytomegalovirus. This test is often used to help diagnose cytomegalovirus infection, especially in individuals with a poorly functioning immune system, such as immunocompromised patients or pregnant women.

 
Principles of the Assay:

The principle of CMV IgM detection is usually based on immunochromatography or enzyme-linked immunosorbent assay (ELISA). These methods use the principle of antibody binding to specific antigens to detect the presence of CMV IgM antibodies in the sample.

In immunochromatography, the sample (usually serum or plasma) is mixed with a reagent containing CMV antigen. If CMV IgM antibodies are present in the sample, they will bind to the antigen to form a complex. This complex will move on the reagent paper or membrane and eventually be detected.

In ELISA, the sample is added to a microplate containing CMV antigen. If CMV IgM antibodies are present in the sample, they will bind to the antigen fixed on the microplate. Then, the presence or absence of CMV IgM is determined by adding an enzyme-labeled secondary antibody that specifically binds to CMV IgM, adding a substrate, and measuring the intensity of the color reaction.

These methods can provide rapid and accurate results to help doctors diagnose CMV infection.

Packaging:
 

Elisa test kits are carefully packaged and shipped in an insulated box to ensure temperature is maintained during transport. The package contains the test kit, instructions, and other supplies needed for the test.

It also includes a return label for customers to return unused portions of the kit after testing is complete. All packages come with tracking information so customers can track the progress of their shipment.
 
 
Storage of test kit:
 

Unopened test kits should be stored at 2-8C upon receipt and the microtiter plate should be kept in a sealed bag to minimize exposure to damp air. Use up the reagents as soon as possible after the kit is unpacked.
 
 
Assay procedure: 
 

• Prepare Reagents: Dilute 1 volume of Washing Buffer (20×)with 19 volumes of distilled water, mix well.
• Add Samples: Open the foil pouch and remove the Microplate. Set up 1 well as Blank, 2 wells as negative control, 2 wells as positive control. After dispensing 100μL of Sample Diluent , dispense 10μL of sample, 100μL negative control or positive control to the respective wells(without Sample Diluent). Gently vibrating the plate.
• Incubate: Cover the Microplate with plate cover and incubate the Microplate in a thermostat-controlled water-bath or microplate incubator at 37℃ for 30 minutes.
• Wash the Plate: Remove the plate cover. Aspirate the contents of all wells. Fill the wells with the diluted washing buffer ( 10~20 seconds to soak) then aspirate again. Repeat the procedure for 5 times. Make sure that the rest volume is minimal, by tapping plate onto absorbent paper.
• Add Conjugate: Add 100μL of conjugate to each well (except the blank well).
• Incubate: Cover the Microplate and incubate the plate at 37℃ for 30 minutes.
• Wash the Plate: Repeat the wash procedure as in step 4.
• Add Substrate: Add 50μL of Substrate Solution A and 50μL of Substrate Solution B to each well, mix well. Cover and incubate at 37℃ for 15 minutes.
• Stop reaction: Add 50μL Stop Solution to each well, mix well.
• Read the absorbance at 450 nm. If a dual wavelength measurement is used, the reference wavelength should be selected from 620nm to 690nm.
