Immune colloidal gold method: using colloidal gold immunochromatography, the reagent contains the anti-hemoglobin antibody which is fixed in the test area on the membrane and the corresponding antibody in the control area. During the test, the diluent of the specimen drops into the test area and chromatographs upward under the capillary effect. If it is positive, the specimen contains hemoglobin and a purple band appears in the test area. If it is negative, there is no hemoglobin in the specimen and there will be no purple band in the test area. Regardless of whether hemoglobin is present in the specimen, a purple-red band will appear in the quality control area. The purple-red band appearing in the quality control area is a standard for judging whether there are enough samples and whether the chromatography process is normal, and also serves as an internal control standard for reagents.

Chemical method: Hemoglobin has peroxidase activity, which can decompose peroxide to release oxygen atoms, which oxidize the tetramethylbenzidine indicator, causing the indicator to change color. When the sample contains a certain amount of human hemoglobin, the color of the test paper will change from orange to yellow-green or dark green, and a very high concentration of hemoglobin will continue to change the color to dark blue.
