In biochemistry, incubators are fundamental for studying enzyme activity and reaction kinetics. Enzymes have precise optimal temperatures and pH ranges at which their catalytic efficiency is maximal. A standard or refrigerated incubator provides the stable, controlled temperature required for these experiments.
For instance, a reaction studying a thermophilic enzyme might be incubated at 65°C, while one for a cold-adapted enzyme might require 4°C. Time-course assays, where samples are withdrawn at intervals to measure product formation, demand that the reaction mixture be held at a constant temperature to ensure meaningful kinetic data (to calculate Vmax, Km). Beyond temperature, some incubators are used to gently mix reaction vessels to ensure homogeneity.
In molecular biology, many core techniques rely on incubation: restriction enzyme digests, ligations, and polymerase chain reaction (PCR) all require precise thermal control, provided by specialized thermal cyclers or water baths. The incubator’s role here is to eliminate temperature as a variable, allowing the biochemist to isolate and study the intrinsic properties of the biomolecule—its structure, function, and interaction with substrates or inhibitors—under reproducible conditions.
