Introduction about incubator
Structural biology efforts to solve the architecture of membrane proteins (like GPCRs or ion channels) often begin with their expression in heterologous systems, a process reliant on incubators.
These proteins are typically expressed in insect or mammalian cells (e.g., using the baculovirus system in insect cells). Culturing these cells at the correct temperature (e.g., 27-28°C for insect cells) and with proper aeration in a shaking incubator is the first critical step to obtain sufficient biomass. After expression, the cells are harvested, and the fragile membrane proteins are extracted with detergents. Subsequent purification steps might require incubation at 4°C in a refrigerated cabinet or cold room to maintain stability.
Finally, the crucial step of crystallization often involves incubating droplets of purified protein with precipitant solutions at a constant, vibration-minimized temperature (e.g., 4°C, 20°C) for days to weeks. Specialized, low-vibration incubators or warm rooms are used for these crystallization trays. A stable thermal environment throughout this multi-week pipeline is essential to produce the high-quality, ordered crystals needed for X-ray diffraction, enabling medicine design targeting these important therapeutic targets.
