39 Introduction about incubator
The generation of stable, clonal animal cell lines—a cornerstone of biopharmaceutical production and functional genetics—is a process critically dependent on incubator performance. After transfecting cells with a gene of interest (e.g., for a therapeutic antibody), a selection agent (like an antibiotic) is added.
The rare cells that have integrated the gene must proliferate from a single cell into a colony. This occurs over 1-3 weeks in a CO2 incubator. The stability of the environment is paramount: pH shifts from CO2 fluctuation can kill fragile, single cells; evaporation from low humidity can concentrate toxic selection agents. Many protocols now use cloning cylinders or limiting dilution in 96-well plates, which are entirely incubated within this controlled environment.
For the highest assurance of clonality, automated cell imagers are often housed inside incubators to monitor single cells without disturbing them. Once colonies form, they are picked and expanded, again within the incubator. The final clonal cell line’s productivity and stability are directly influenced by the consistency of conditions during this vulnerable selection and expansion phase. A high-quality, stable incubator is therefore the unsung hero in developing the cell lines that produce blockbuster biologic drugs.
